A Multiomics Approach for Investigating the Heterogeneity of Memory T Cells


The post-thymic activation of T cells involves a heterogeneous population of cells at different stages of differentiation, including naïve, stem cell memory, central memory, effector memory and terminal effector T cells. These different T cell subsets are themselves highly heterogeneous, and their identification and characterization require not one or a few but, rather, a combination of several markers. Researchers are now seeking new markers that might provide insights into the T cell differentiation and development pathway and to potentially identify new cell populations. These exploratory studies require development of multiomics approaches that combine high-dimensional flow cytometry and single-cell RNA sequencing (scRNA-Seq) for phenotypic and genomic analyses at the single-cell level.

 

Here, we share a data sheet that presents a workflow to identify markers for deeper characterization of immune cells, with data on a 9-color CD8+ T cell panel and a 16-color CD8+ and CD4+ T cell panel on the BD LSRFortessa™ X-20 Cell Analyzer.
